APVO210 is a bispecific fusion protein therapeutic developed using Aptevo Therapeutics' ADAPTIR platform. It consists of an anti-CD86 antibody fused to a modified form of interleukin-10 (IL-10). The drug is designed to selectively deliver IL-10 to CD86-positive antigen-presenting cells (such as monocytes, macrophages, and dendritic cells), thereby inducing immunosuppressive signaling specifically in these cells without activating T or B lymphocytes. This targeted approach aims to promote the differentiation of tolerogenic dendritic cells and the induction of antigen-specific regulatory T cells (Tr1), leading to immune tolerance while minimizing systemic immunosuppression and associated toxicities seen with other IL-10 therapies. APVO210 was investigated for autoimmune and inflammatory diseases such as ulcerative colitis and psoriasis but its development was discontinued due to issues with anti-drug antibodies[1][3][4][5][6][7][8].
