Overview

APVO455 is a preclinical, bispecific T-cell engager targeting Nectin-4 and CD3. It is designed to stimulate anti-tumor immunity by redirecting T cells to kill Nectin-4 positive tumor cells. Nectin-4 is highly expressed in several solid tumors, including bladder, breast, non-small cell lung cancer (NSCLC), and head and neck cancers. The molecule deploys a proprietary CD3 binding domain optimized for reduced cytokine release, aiming to minimize systemic toxicity and adverse events—a critical limitation for T-cell engagers, particularly in solid tumor indications. APVO455 is developed on Aptevo’s ADAPTIR and ADAPTIR-FLEX bispecific platform technologies[1][3][4][5][10].
