APVO711 is a bispecific antibody developed by Aptevo Therapeutics, designed as a dual-mechanism immunotherapy for cancer. It simultaneously blocks the PD-1/PD-L1 immune checkpoint pathway and activates CD40 on antigen-presenting cells. This approach aims to both relieve immune suppression (via PD-L1 inhibition) and enhance T-cell priming (via CD40 activation), potentially resulting in more robust anti-tumor responses, especially in solid tumors resistant to standard checkpoint inhibitors. Unlike first-generation CD40 monoclonal antibodies, APVO711’s design restricts CD40 activation to the tumor microenvironment by requiring crosslinking through PD-L1 binding, which may reduce systemic toxicity. The molecule is currently in preclinical development and leverages Aptevo’s proprietary ADAPTIR platform[1][3][5][6].
