Overview

APX005M + nivolumab is a combination of two monoclonal antibodies used in cancer immunotherapy. APX005M (also known as sotigalimab) is a humanized IgG1 agonistic monoclonal antibody that targets CD40, a co-stimulatory receptor on antigen-presenting cells. By activating CD40, APX005M enhances the activation of both innate and adaptive immune responses against tumors. Nivolumab is a humanized IgG4 monoclonal antibody that inhibits programmed cell death protein 1 (PD-1), thereby blocking an immune checkpoint and promoting T-cell-mediated anti-tumor activity. The combination aims to overcome resistance to PD-1 inhibitors by synergistically activating the immune system through complementary mechanisms. This regimen has been studied primarily in patients with advanced or metastatic melanoma who have progressed on prior anti-PD-1 therapy[1][8][6].
