APX2009 is a second-generation small molecule inhibitor targeting the redox activity of apurinic/apyrimidinic endonuclease 1/redox factor-1 (APE1/REF-1), a multifunctional protein involved in DNA base excision repair and redox regulation of transcription factors. By inhibiting APE1/REF-1, APX2009 disrupts cellular signaling pathways that promote tumor cell survival, proliferation, migration, invasion, angiogenesis, and metastasis. Preclinical studies have demonstrated its anticancer properties in colon, bladder, pancreatic, prostate cancer models as well as breast cancer cell lines. Additionally, APX2009 exhibits antiangiogenic effects relevant to ocular diseases such as wet age-related macular degeneration and diabetic macular edema by blocking endothelial cell proliferation and neovascularization[1][2][6][7].
