APX2039 is an investigational, orally active small-molecule antifungal agent that potently inhibits the fungal Gwt1 enzyme, which catalyzes an early step in glycosylphosphatidylinositol (GPI) anchor biosynthesis required for anchoring mannoproteins to the fungal cell wall.[1][3][5][7] By selectively targeting fungal Gwt1 (without inhibiting the closest human homolog PIGW), APX2039 disrupts cell-wall integrity and is fungicidal against Cryptococcus neoformans and related pathogens, achieving rapid clearance of fungal burden in mouse and rabbit models of cryptococcal meningitis with excellent central nervous system penetration.[3][5][7] APX2039 is part of the “gepix” class of Gwt1 inhibitors being advanced by Amplyx Pharmaceuticals (acquired by Pfizer) as a next-generation oral monotherapy candidate for cryptococcal meningitis and other invasive fungal infections.[3][5][7][11][12]
