Overview

APX2048 is a preclinical-stage small molecule inhibitor developed by Apexian Pharmaceuticals. It is a second-generation analog of APX3330, specifically designed to target the redox signaling function of APE1/Ref-1 (Apurinic/apyrimidinic endonuclease 1/Redox effector factor 1). APE1/Ref-1 is a multifunctional protein; while its endonuclease activity is involved in DNA base excision repair, its redox activity facilitates the reduction and activation of transcription factors such as NF-κB, AP-1, and HIF-1α. These factors are critical drivers of inflammation, angiogenesis, and tumor progression. By selectively inhibiting the redox function of Ref-1, APX2048 aims to disrupt these pathological signaling pathways. The compound is being explored for potential applications in oncology and the treatment of vasoproliferative retinal diseases.
