Overview

APX3448 is a small molecule inhibitor developed by Apexian Pharmaceuticals that targets the redox signaling function of Apurinic/apyrimidinic endonuclease 1/Redox effector factor 1 (APE1/Ref-1). Unlike traditional inhibitors that target the DNA repair (endonuclease) function of APE1, APX3448 specifically blocks the protein's ability to reduce and activate key transcription factors, including NF-κB, AP-1, HIF-1α, and STAT3. These factors are critical drivers of tumor growth, survival, and angiogenesis. As a second-generation compound in Apexian's pipeline, APX3448 is designed to offer improved potency and pharmacological properties compared to the lead candidate APX3330. It is primarily being investigated for the treatment of advanced solid tumors and other conditions where APE1/Ref-1 signaling is dysregulated.
