APX3558 is a second-generation small molecule inhibitor developed by Apexian Pharmaceuticals. It specifically targets the redox signaling function of Apurinic/apyrimidinic endonuclease 1/Redox effector factor 1 (APE1/Ref-1). APE1/Ref-1 is a dual-function protein involved in both DNA base excision repair and the reduction-oxidation (redox) regulation of transcription factors. APX3558 is designed to inhibit the redox function, which in turn modulates the activity of several transcription factors critical for tumor growth, survival, and inflammation, including NF-κB, AP-1, and HIF-1α. As a more potent analog of the lead candidate APX3330, APX3558 is being explored for its potential in treating various advanced solid tumors and inflammatory conditions where APE1/Ref-1 signaling is dysregulated.
