APX4531 is a small molecule inhibitor developed by Apexian Pharmaceuticals that targets the redox signaling function of the Apurinic/apyrimidinic endonuclease 1/Redox effector factor 1 (APE1/Ref-1) protein. It is a second-generation analog of the company's lead compound, APX3330. APX4531 specifically inhibits the redox-mediated activation of several transcription factors, including NF-kB, AP-1, HIF-1α, and STAT3, which are known to drive tumor progression, survival, and inflammatory responses. By selectively targeting the redox function while sparing the DNA repair (endonuclease) activity of APE1/Ref-1, APX4531 aims to provide a more targeted therapeutic approach for advanced solid tumors and various inflammatory or neurodegenerative conditions.
