APX601 is a novel anti-TNFR2 antagonist monoclonal antibody developed for cancer immunotherapy. Its mechanism of action involves targeting and antagonizing tumor necrosis factor receptor 2 (TNFR2), which is highly expressed on regulatory T cells (Tregs) and certain tumor cells within the tumor microenvironment. By blocking TNFR2, APX601 depletes and inactivates TNFR2-expressing Tregs, reverses myeloid-mediated T cell suppression, and directly kills TNFR2-expressing tumor cells. This leads to reversal of immune suppression in the tumor microenvironment and unleashes immune-mediated anti-tumor activity. Preclinical studies have demonstrated potent anti-tumor effects across multiple solid tumor models[1][6][8].
