AQ128 is a highly potent, selective small-molecule inhibitor of transforming growth factor-β-activated kinase 1 (TAK1) under development by Aqilion. TAK1 is a central signaling hub that integrates innate immune responses and inflammasome activation, regulating key pro-inflammatory cytokines such as TNF-alpha, IL-1β, and IL-6 via NF-kB and JNK signaling pathways. AQ128 is being developed as a topical treatment for psoriasis, aiming to provide a first-in-class therapeutic option that matches the efficacy of potent corticosteroids (like clobetasol) without their associated systemic side effects. The program was previously part of a strategic collaboration with Merck KGaA, which was terminated in 2024, after which Aqilion resumed independent development of the asset.
