Drug intelligence / Profile preview
AqT-Calicheamicin is a specialized antibody-drug conjugate (ADC) payload developed by AqueaTether Therapeutics, a subsidiary or partner of CellMosaic. It consists of the potent cytotoxic agent calicheamicin modified with AqueaTether (AqT) technology, which utilizes a highly hydrophilic, non-immunogenic polymer to improve the solubility and conjugation properties of the payload. Calicheamicin is a small molecule enediyne antibiotic that binds to the minor groove of DNA and undergoes a Bergman cyclization to generate a diradical, leading to site-specific double-strand DNA cleavage and subsequent cell death. The AqT-Calicheamicin asset is primarily used as a platform tool for the development of novel ADCs targeting various cancers, aiming to overcome the hydrophobicity and aggregation issues often associated with traditional calicheamicin-based conjugates.
