Overview

AqT-duocarmycin is a specialized antibody-drug conjugate (ADC) payload developed by AqueaTether Therapeutics, a partner of CellMosaic. It utilizes the proprietary AqueaTether (AqT) technology to modify a duocarmycin analog, a potent cytotoxic small molecule. Duocarmycins function as DNA minor-groove alkylators; they bind specifically to the minor groove of DNA and alkylate the N3 position of adenine, leading to irreversible DNA damage, cell cycle arrest, and apoptosis. The AqT modification is designed to enhance the hydrophilicity and conjugation efficiency of the typically hydrophobic duocarmycin payload, allowing for the development of ADCs with improved pharmacological profiles. It is currently utilized as a platform asset for the construction of novel cancer therapeutics.
