AqT-MMAE is a modified form of the potent tubulin inhibitor monomethyl auristatin E (MMAE), developed by AqueaTether Therapeutics and CellMosaic. It utilizes the AqueaTether (AqT) platform, which incorporates super-hydrophilic linkers derived from sugar alcohols to improve the water solubility and reduce the aggregation of the highly hydrophobic MMAE payload. This technology is designed to enable higher drug-to-antibody ratios (DAR) in antibody-drug conjugates (ADCs) while maintaining stability and biocompatibility. Upon reaching the target cancer cell, the MMAE is released via a cleavable ester bond to exert its antimitotic effect by inhibiting tubulin polymerization, leading to cell cycle arrest and apoptosis.
