AqT-PBD Dimer is a pyrrolobenzodiazepine (PBD) dimer payload developed by AqueaTether Therapeutics (a CellMosaic company) for use in antibody-drug conjugates (ADCs). It utilizes the proprietary AqueaTether (AqT) technology, which incorporates a highly hydrophilic, non-immunogenic, and biodegradable tether to improve the solubility and conjugation efficiency of hydrophobic payloads. The PBD dimer mechanism involves binding to the minor groove of DNA and forming interstrand crosslinks, which leads to DNA damage, inhibition of replication, and subsequent cell death. This payload is intended for the treatment of various cancers when conjugated to a targeting antibody.
