AqT-SN38

AqT-SN38 is a specialized antibody-drug conjugate (ADC) payload developed by AqueaTether Therapeutics, a company associated with CellMosaic. It consists of SN38, the potent active metabolite of the topoisomerase I inhibitor irinotecan, conjugated to the proprietary AqueaTether (AqT) linker technology. The AqT platform utilizes a highly hydrophilic, non-immunogenic polymer tether designed to improve the solubility, stability, and pharmacokinetics of the resulting ADC by masking the hydrophobicity of the payload. By inhibiting topoisomerase I, SN38 stabilizes the covalent complex between the enzyme and DNA, preventing DNA ligation and inducing lethal double-stranded DNA breaks during the S-phase of the cell cycle. AqT-SN38 is primarily utilized as a platform asset for the construction of novel ADCs targeting various solid tumors.
