AQX-1000 is a series of small molecule modulators of Src homology 2 domain-containing inositol 5-phosphatase 1 (SHIP1) developed by Aquinox Pharmaceuticals. SHIP1 is a signaling enzyme primarily expressed in hematopoietic cells that acts as a negative regulator of the PI3K/Akt pathway by converting PI(3,4,5)P3 to PI(3,4)P2. The AQX-1000 program was designed to leverage this mechanism for the treatment of inflammatory diseases and hematologic malignancies. While Aquinox's lead candidate, rosiptor (AQX-1125), was a SHIP1 activator, the AQX-1000 series represented next-generation compounds within their preclinical pipeline intended to address a broader range of indications including heme malignancies.
