AR-12286 + travoprost is a fixed-dose combination of two ophthalmic drugs developed for the reduction of intraocular pressure (IOP) in patients with glaucoma or ocular hypertension. AR-12286 (also known as verosudil) is a highly selective Rho-associated protein kinase (ROCK) inhibitor that lowers IOP primarily by increasing aqueous humor outflow through the trabecular meshwork, reversing pathological changes and improving drainage[1][3][7][8]. Travoprost is a prostaglandin analog prodrug that acts as a full agonist at the prostaglandin F2-alpha receptor, increasing uveoscleral and trabecular outflow to reduce IOP[6]. The combination targets complementary pathways to enhance IOP lowering efficacy. AR‑12286 was developed by Aerie Pharmaceuticals and has demonstrated significant efficacy in clinical trials for primary open-angle glaucoma and ocular hypertension[5][7].
