Overview

AR-678 is a selective small molecule inhibitor of **Checkpoint kinase 1 (Chk1)**, a critical protein in the DNA damage response (DDR) pathway. Developed by **Array BioPharma** (now part of **Pfizer**), AR-678 targets the S-phase and G2/M checkpoints. In tumor cells exhibiting high levels of endogenous replicative stress, such as certain melanomas, Chk1 inhibition prevents the repair of DNA damage and forces the cells into premature mitosis, leading to mitotic catastrophe and apoptosis. Preclinical studies have demonstrated that AR-678 possesses potent single-agent cytotoxic activity in melanoma cell lines with nanomolar IC50 values, while exhibiting significantly lower toxicity in normal cells. This selectivity suggests that markers of DNA damage could serve as predictive biomarkers for sensitivity to AR-678 and similar Chk1 inhibitors.
