**AR-701** is a cocktail of two fully human IgG1 monoclonal antibodies (mAbs), AR-703 and AR-720, discovered from convalescent COVID-19 patients by Aridis Pharmaceuticals. AR-703 inhibits viral fusion and entry into human cells by binding to the S2 stalk region of betacoronavirus spike proteins, while AR-720 blocks viral binding to the human ACE2 receptor by targeting the receptor binding domain (RBD) of the SARS-CoV-2 spike protein. The combination exhibits synergistic activity with broad pan-coronavirus neutralization, including SARS-CoV-2 variants (Alpha, Beta, Gamma, Delta, Epsilon, Omicron subvariants BA.1/BA.2/BA.4/BA.5), SARS, MERS, and common cold coronaviruses. It demonstrated complete protection in non-human primate models of COVID-19 via inhaled or intramuscular administration, reducing viral load in lungs, nasal sinus, and oropharynx, and is being developed for prophylactic and therapeutic use in non-hospitalized patients, with formulations for self-administered inhalation via nebulizer or long-acting intramuscular injection.[1][7][10]
