**AR-9.6** is a novel monoclonal antibody (mAb) that specifically binds to the SEA domain 5 on MUC16, a mucin protein aberrantly glycosylated in cancers, interfering with its interaction with ErbB (EGF) receptors on tumor cell surfaces. This binding abrogates downstream oncogenic signaling pathways, including AKT and GSK3β, thereby inhibiting pro-oncogenic activity and conferring a therapeutic response in MUC16-expressing tumors. Initially developed at the University of Nebraska, it has been licensed to Quest PharmaTech (through subsidiary AmrutBio) and OncoCare Therapeutics for further development as a theranostic agent, with preclinical validation demonstrating strong tumor targeting in ovarian and pancreatic cancer xenograft models via immunoPET imaging using 89Zr-labeled conjugates.[1][3][5]
