Overview

AR-C78511 is a potent, small molecule inverse agonist of the P2Y12 receptor, a G protein-coupled receptor (GPCR) primarily expressed on platelets that plays a critical role in ADP-induced aggregation. Developed by AstraZeneca, AR-C78511 is distinguished from traditional P2Y12 antagonists, such as cangrelor, by its ability to suppress the constitutive (basal) signaling activity of the receptor. This property is particularly relevant in the context of type 2 diabetes mellitus, where P2Y12 receptors are often overexpressed and hyperactivated. Research indicates that AR-C78511 exhibits superior antiplatelet and antithrombotic efficacy in diabetic models compared to standard antagonists, suggesting that targeting constitutive P2Y12 activity may be a viable strategy for managing platelet hyperactivity in metabolic diseases. It is primarily used as a pharmacological research tool to investigate the role of P2Y12 in cardiovascular complications.
