Overview

AR20.5 is a murine monoclonal antibody (IgG1 subtype) that specifically binds with high affinity to tumor-associated antigen MUC1, recognizing the tandem repeat peptide sequence DTRPAP in the MUC1 extracellular domain. MUC1 is aberrantly under-glycosylated in cancer cells, which exposes epitopes targeted by AR20.5, allowing for selective binding to malignant versus normal tissues. AR20.5 has been evaluated for both therapeutic and diagnostic applications: as a monotherapy to induce MUC1-specific immune responses, as a cancer vaccine component, and as an immunoPET imaging probe (when conjugated to radioisotopes, e.g. zirconium-89), detecting MUC1-expressing tumors in preclinical models. While AR20.5 has shown immune activation and tumor growth suppression in mice, clinical efficacy remains unproven. The antibody functions partly by forming immune complexes with circulating MUC1 or MUC1-expressing tumor cells, inducing immune responses[1][2][3][6].
