Overview

AR234960 is a potent and selective small molecule agonist of the Mas receptor (Mas1), a G protein-coupled receptor that serves as the primary functional receptor for the protective arm of the renin-angiotensin system (RAS). Originally identified by Arena Pharmaceuticals (now part of Pfizer), AR234960 is used to investigate the therapeutic potential of Mas1 activation in cardiovascular and pulmonary disorders. In preclinical models of pulmonary arterial hypertension (PAH), such as the Sugen-Hypoxia mouse model, AR234960 has been shown to mimic the beneficial effects of angiotensin-converting enzyme 2 (ACE2) by reducing pulmonary arterial pressure and vascular remodeling. The compound's mechanism of action involves the rescue of cytoskeletal and microtubule defects, which are critical for maintaining vascular integrity. Unlike angiotensin-(1-7), which was historically considered the endogenous ligand for Mas1, AR234960 provides a direct and stable means of receptor activation, making it a significant lead for the development of small molecule therapies for PAH.
