Drug intelligence / Profile preview
Ara h 6 (Arachis hypogaea allergen 6) is a naturally occurring 2S albumin peanut allergen protein being investigated as a candidate component for improved allergen immunotherapy (AIT). Developed primarily in preclinical translational research at Amsterdam UMC, Ara h 6 is a purified native protein used as an active ingredient in experimental allergen immunotherapy formulations. It shares approximately 60% sequence identity and multiple IgE-binding epitopes with the dominant peanut allergen Ara h 2, making it one of the most diagnostically and immunologically significant peanut allergens. Ara h 6 targets the IgE-mediated immune response, specifically binding to IgE antibodies to modulate downstream effector cells (such as mast cells and basophils) involved in allergic sensitization. In preclinical models, sublingual immunotherapy (SLIT) with purified Ara h 6 has been shown to protect against subsequent sensitization and drive the expansion of FoxP3+Helios− regulatory T cells (Tregs), suggesting a tolerogenic mechanism of action. It is currently being screened alongside other candidate components (including modified allergens, nanoparticles, adjuvants, and plant-based bioparticles) using patient-derived PBMCs and serum to develop safer and more effective vaccines for peanut allergy, food allergy, and other IgE-mediated allergic conditions.
