Overview
Araris anti-HER2 ADC is a preclinical antibody-drug conjugate (ADC) developed by Araris Biotech, a subsidiary of Taiho Pharmaceutical. The drug targets Human Epidermal Growth Factor Receptor 2 (HER2) and utilizes the proprietary AraLinQ site-specific conjugation technology. It features a unique dual-payload design with a drug-to-antibody ratio (DAR) of 4 (2+2), incorporating two different exatecan-based topoisomerase-1 inhibitor (TOP1i) payloads. One payload is designed for high intracellular accumulation and potency, while the other possesses bystander activity to address tumor heterogeneity. In proof-of-concept studies, trastuzumab was employed as the targeting antibody. Preclinical evaluations have shown the ADC to have superior anti-tumor efficacy compared to trastuzumab deruxtecan (Enhertu) in models of HER2-expressing solid tumors, including breast cancer.
