Overview
Araris anti-Nectin-4 ADC is an antibody-drug conjugate (ADC) being developed by Araris Biotech AG. It targets Nectin-4, a tumor-associated antigen overexpressed in various solid tumors, including urothelial carcinoma and triple-negative breast cancer (TNBC). The ADC utilizes the antibody enfortumab conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) through a proprietary site-specific peptide linker technology. This linker technology is designed to improve the therapeutic index by enhancing stability and payload delivery. Preclinical data suggests superior efficacy and tolerability compared to the approved Nectin-4 ADC, enfortumab vedotin. Additionally, Araris is developing a version of this ADC featuring a triple-warhead payload that combines MMAE with topoisomerase-1 inhibitors.
