Overview
ARAspheres is a preclinical-stage radiopharmaceutical developed by BlueWave Therapeutics for the treatment of glioblastoma. The technology utilizes alginate-based biopolymer nanospheres as a delivery vehicle for Actinium-225 (225Ac), a potent alpha-emitting radioisotope. These nanospheres are surface-modified with RGD (arginine-glycine-aspartic acid) peptides to target integrin receptors, specifically alpha-V beta-3 (αvβ3), which are highly expressed on glioblastoma cells and tumor-associated blood vessels. Upon binding and internalization, the Actinium-225 delivers high-LET (linear energy transfer) alpha radiation, causing irreparable double-strand DNA breaks within the target cells. This localized radiation therapy aims to maximize tumor cell killing while sparing adjacent healthy brain tissue.
