**ARB-1740** is a second-generation, clinical-stage RNA interference (RNAi) therapeutic developed by Arbutus Biopharma for chronic hepatitis B (CHB) infection. It consists of three small interfering RNAs (siRNAs) formulated in lipid nanoparticles (LNP) for targeted hepatic delivery, enabling pan-genotypic silencing of HBV antigens including HBsAg, HBeAg, HBcAg, and HBV genome transcripts via post-transcriptional gene silencing. Preclinical studies demonstrated potent reductions in HBV viremia (2.3 log10), HBsAg (2.6 log10), and HDV viremia (1.6 log10) in humanized mouse models; it advanced to Phase II trials but was discontinued due to suboptimal efficacy.[1][2][3][4][5]
