Overview

ARB011 is an allogeneic, non-viral, RNA-based "Flash" CAR-NK cell therapy targeting Cadherin-17 (CDH17), an oncofetal protein overexpressed in gastrointestinal (GI) cancers. Developed by ArbeleBio, the therapy is produced by electroporating ex vivo expanded human peripheral blood natural killer (NK) cells with mRNA encoding a humanized anti-CDH17 single-chain variable fragment (scFv) fused with transactivation domains. This approach allows for an "off-the-shelf" treatment that avoids the risks associated with viral vectors and permanent genomic integration. Preclinical studies have demonstrated significant CDH17-specific cytotoxicity against GI cancer cells, accompanied by the release of IFN-γ and TNF-α, suggesting potential efficacy in treating advanced gastrointestinal malignancies.
