Overview

Arbaprostil is a synthetic analog of prostaglandin E2 (PGE2), specifically the 15(R)-methyl derivative. Developed by Upjohn, it acts as a gastric cytoprotective agent and an inhibitor of gastric acid secretion. It was primarily investigated for the prevention and healing of gastric mucosal damage and ulcers associated with the use of nonsteroidal anti-inflammatory drugs (NSAIDs) in patients with conditions like rheumatoid arthritis and osteoarthritis. Clinical trials demonstrated its efficacy in resolving gastric lesions, though its use was associated with dose-dependent diarrhea. Development was eventually discontinued in favor of other prostaglandin analogs like misoprostol.
