ARC-520 + tenofovir is a combination regimen used in the treatment of chronic hepatitis B virus (HBV) infection. ARC-520 is an RNA interference (RNAi) therapeutic comprising small interfering RNAs (siRNAs) that target HBV viral transcripts derived from covalently closed circular DNA (cccDNA), resulting in the reduction of hepatitis B surface antigen (HBsAg) and other viral antigens. Tenofovir is a nucleoside analogue reverse transcriptase inhibitor (NUC) that inhibits HBV DNA polymerase, thereby suppressing HBV DNA replication. In clinical studies, administration of ARC-520 in combination with tenofovir led to significant reductions in HBsAg and HBeAg levels and sustained suppression of HBV DNA. The combination regimen was generally well tolerated with no severe safety signals observed[3][4][5][7][8][9].
