ARC-520 + tenofovir disoproxil + pegylated interferon alpha 2a is a combination regimen investigated for the treatment of chronic hepatitis B virus (HBV) infection. ARC-520 is an RNA interference (RNAi) therapeutic designed to target and degrade hepatitis B viral mRNA transcripts produced from covalently closed circular DNA (cccDNA), thereby reducing the expression of viral proteins such as HBsAg and core-related antigens. Tenofovir disoproxil is a nucleotide reverse transcriptase inhibitor, which blocks the enzyme needed for viral DNA replication. Pegylated interferon alpha 2a is an immunomodulatory cytokine that stimulates antiviral immune responses and can promote seroclearance of viral antigens. The combination aims to reduce viral antigens, suppress viral replication, and enhance host immune control to increase the rates of functional cure (seroclearance of HBsAg). This regimen has been studied in multi-center, open-label clinical trials for both HBeAg-positive and HBeAg-negative chronic HBV patients[1][5][6][3][2].
