**ARC-SparX** is Arcellx's controllable CAR-T cell therapy platform composed of two coordinated biologic components: autologous universal ARC-T cells and separately administered soluble SparX proteins that bridge the ARC-T cells to tumor-associated antigens. The system uses Arcellx's D-Domain synthetic binding scaffold and is designed so that ARC-T cells become activated only when a SparX protein is bound to a target cell, enabling on/off control through SparX dosing and potential retargeting across different antigens. Publicly described programs built on this platform include ACLX-001 targeting BCMA in relapsed or refractory multiple myeloma and ACLX-002 targeting CD123 in relapsed or refractory acute myeloid leukemia and high-risk myelodysplastic syndrome; Kite has exercised its option to license ACLX-001.
