Overview

ARC031 is a small molecule derivative of nilvadipine (ARC029) developed by Archer Pharmaceuticals for the treatment of Alzheimer's disease. It was specifically engineered to retain the amyloid-beta lowering properties of its parent compound while eliminating the vasodilatory effects associated with calcium channel blockade. By reducing the levels of soluble amyloid-beta proteins in the brain, ARC031 aims to prevent the neuronal degeneration characteristic of Alzheimer's. The lack of blood pressure-lowering activity was intended to allow for higher therapeutic doses without the risk of hypotension. The drug was manufactured to GMP standards and evaluated in Phase I clinical trials using both immediate-release and controlled-release oral formulations, although development activity has not been publicly reported since 2016.
