arcotatug tavatecan

Arcotatug tavatecan (IBI343) is a next-generation antibody-drug conjugate (ADC) developed by Innovent Biologics. It consists of a humanized monoclonal antibody targeting Claudin 18.2 (CLDN18.2), conjugated to exatecan, a topoisomerase I inhibitor payload via a cleavable linker[1][6]. The drug is designed to selectively deliver the cytotoxic agent to CLDN18.2-expressing tumor cells, leading to DNA damage and cell death primarily in cancer cells overexpressing this target[1][6]. Arcotatug tavatecan is being investigated for the treatment of advanced solid tumors with high CLDN18.2 expression, including gastric cancer, gastroesophageal junction adenocarcinoma, pancreatic ductal adenocarcinoma (PDAC), biliary tract cancer (BTC), colorectal cancer, mucosal melanoma, and non-small cell lung cancer[6]. Early clinical data have shown promising objective response rates and disease control rates in patients with advanced PDAC and BTC expressing CLDN18.2[6].
