ARCT-021 is a self-amplifying mRNA (sa-mRNA) vaccine candidate developed by Arcturus Therapeutics for the prevention of COVID-19. It utilizes the STARR self-replicating mRNA technology combined with LUNAR lipid nanoparticle delivery to enhance and prolong antigen expression, enabling effective vaccination at lower doses. The vaccine encodes both the replication complex of Venezuelan equine encephalitis virus (VEEV) and the SARS-CoV-2 spike protein, targeting myocytes and dendritic cells to induce robust humoral and cellular immune responses. Clinical trials demonstrated that ARCT-021 was immunogenic with a favorable safety profile in both younger and older adults, eliciting neutralizing antibodies as well as CD4+ Th1-biased T-cell responses. Despite promising early results, development for COVID-19 was discontinued after Phase 2 trials[1][2][3][4][5][6][7].
