ARCT-154 is a self-amplifying mRNA (sa-mRNA) vaccine developed for the prevention of COVID‑19. It encodes the full-length spike protein of SARS-CoV‑2, specifically the D614G variant, using an sa-mRNA platform derived from Venezuelan equine encephalitis virus (VEEV), with structural genes replaced by those encoding the spike protein. The vaccine is delivered via lipid nanoparticles and is designed to induce robust and durable immune responses at lower doses compared to conventional mRNA vaccines. Clinical studies have shown that ARCT‑154 elicits strong immunogenicity against both ancestral and Omicron variants of SARS-CoV‑2, with efficacy demonstrated in multiple age groups. The vaccine was developed by Arcturus Therapeutics in collaboration with Vinbiocare Biotechnology and Meiji Seika Pharma, and has been registered or recommended for approval in several regions including Japan, Vietnam, Iceland, Norway, Liechtenstein, and the European Union[1][3][5][8].
