ARCT-165 is a next-generation self-amplifying mRNA (sa-mRNA) vaccine candidate developed by Arcturus Therapeutics for the prevention of COVID-19. Utilizing the company's proprietary STARR™ (Self-Transcribing and Replicating RNA) technology, ARCT-165 encodes the SARS-CoV-2 spike protein and includes a viral replicase that allows the mRNA to replicate itself within the host cell. This mechanism enables sustained antigen expression at significantly lower doses compared to conventional mRNA vaccines. The vaccine is delivered using the LUNAR® lipid nanoparticle platform, which facilitates efficient intracellular delivery. ARCT-165 was designed to target SARS-CoV-2 variants of concern and has been evaluated in Phase 1/2 clinical trials as both a primary vaccination series and a booster dose.
