Drug intelligence / Profile preview
ARD-100 is an oral, clinical-stage monomethyl fumarate (MMF) prodrug being developed by Ardanza Biopharma (in partnership with Cellix Bio) for the treatment of relapsing-remitting multiple sclerosis (RRMS). Chemically, ARD-100 consists of MMF linked to eicosapentaenoic acid (EPA) via a glycerin spacer. This design allows for gradual esterase-mediated cleavage during intestinal transit, delaying MMF release and reducing the peak plasma concentration (Cmax) while maintaining the daily area under the curve (AUC). This peak-sparing pharmacokinetic profile is designed to improve gastrointestinal tolerability and reduce flushing compared to dimethyl fumarate (DMF) and diroximel fumarate (DRF), thereby enhancing patient adherence.
