Drug intelligence / Profile preview
ARD-489AD is an orally bioavailable, highly selective small molecule inhibitor of Tyrosine Kinase 2 (TYK2), currently under development by Khui Smart Medicine (科辉智药). TYK2 is a member of the Janus kinase (JAK) family that mediates signaling for cytokines such as IL-23, IL-12, and Type I interferons, which are central to the pathogenesis of various autoimmune and inflammatory diseases. ARD-489AD is designed as an allosteric inhibitor, likely targeting the JH2 pseudokinase domain of TYK2 to achieve high selectivity over other JAK family members (JAK1, JAK2, and JAK3), thereby potentially reducing off-target effects such as hematological toxicity or lipid imbalances. The drug is currently being evaluated in Phase 1 clinical trials in China to assess its safety, tolerability, and pharmacokinetics in healthy adult volunteers.
