Overview

Ardipusilloside I is a **natural triterpenoid saponin** extracted from the plant *Ardisia pusilla* A.DC, belonging to the family Myrsinaceae. It exhibits significant effects on gastrointestinal motility by stimulating smooth muscle contractility, enhancing phosphorylation of the 20 kDa myosin light chain (MLC20) via **activation of myosin light chain kinase (MLCK)**, and increasing myosin Mg²⁺-ATPase activity. Ardipusilloside I has demonstrated benefits in both normal and constipation models in rodents, indicating potential as a therapeutic for constipation. Additionally, it shows **potent antitumor activity** against various tumor types (liver, stomach, ovarian, lung, and glioma) in both in vitro and in vivo models. Its antitumor mechanism is believed to involve effects on cancer cell proliferation, invasion, metastasis, and possibly immunomodulation. Poor gastrointestinal absorption and risk of hemolysis limit conventional oral or intravenous administration, but local delivery approaches (e.g., polymer microspheres) are being explored[1][2][3].
