Overview

Arginase is a manganese-dependent metalloenzyme of the urea cycle that catalyzes the hydrolysis of L-arginine to L-ornithine and urea. As a therapeutic agent, arginase is developed in two primary clinical contexts: as an enzyme replacement therapy (such as pegzilarginase) to treat arginase 1 deficiency (hyperargininemia), and as an arginine-depleting antineoplastic therapy (such as PEG-BCT-100) to treat arginine-auxotrophic cancers (including hepatocellular carcinoma, acute myeloid leukemia, and other solid tumors). By depleting systemic L-arginine, therapeutic arginase starves cancer cells that lack the ability to synthesize arginine de novo due to deficient expression of argininosuccinate synthetase (ASS1) or ornithine transcarbamylase (OTC).
