A cancer vaccine composed of peptides derived from the enzyme arginase-1 (ARG1), designed to stimulate an immune response against cells expressing arginase-1. Arginase-1-producing cells in the tumor microenvironment suppress T cell-mediated anti-tumor responses by depleting L-arginine, an amino acid essential for T cell function. The activation of ARG1-specific T cells through vaccination aims to eliminate or reprogram these immunosuppressive cells, restore L-arginine levels, and enhance local Th1 inflammation, thereby improving anti-tumor immunity. In clinical studies, the vaccine consists of three 20-mer peptides from immunogenic regions of ARG1 and is administered with Montanide ISA-51 as an adjuvant. It has shown safety and immunogenicity in phase I trials involving patients with treatment-refractory metastatic solid tumors[2][3][5].
