Overview

Arginyl-glycyl-aspartic acid (RGD) is a tripeptide sequence that serves as a minimal recognition motif for several integrin receptors, including $\alpha_v\beta_3$, $\alpha_v\beta_5$, and $\alpha_5\beta_1$. These receptors are frequently overexpressed on the surface of cancer cells and activated endothelial cells during angiogenesis, as well as in fibrotic tissues. Consequently, RGD peptides are widely employed as targeting moieties in the design of "smart" drug delivery systems, such as nanoparticles, extracellular vesicles, and radiopharmaceuticals, to enhance the localization of therapeutic or diagnostic agents to specific pathological sites. The motif is found in various extracellular matrix proteins like fibronectin and vitronectin, where it mediates cell-matrix interactions.
