Overview

ARGX-101 (also known as TSP-101) is a preclinical-stage monoclonal antibody developed by argenx that targets the fibroblast growth factor-inducible 14 (Fn14) receptor, a member of the tumor necrosis factor receptor superfamily (TNFRSF12A). Fn14 is the sole signaling receptor for the cytokine TWEAK (TNF-like weak inducer of apoptosis). The TWEAK/Fn14 axis is a key regulator of tissue remodeling and inflammation, and its overactivation is linked to chronic inflammatory diseases, fibrosis, and the progression of various solid tumors. ARGX-101 is designed to bind Fn14 and block its interaction with TWEAK, thereby inhibiting downstream pro-inflammatory and pro-survival signaling pathways. The program was the first to emerge from argenx's Target Seed Program (TSP) but is not currently listed in the company's active clinical pipeline, indicating it is either in early development or a legacy asset.
