Overview

ARGX-111 is a human monoclonal antibody developed to target c-Met (also known as the mesenchymal-epithelial transition factor), a receptor tyrosine kinase associated with tumor growth, metastasis, and poor prognosis in various cancers. The drug is engineered using argenx's SIMPLE Antibody™ platform and incorporates NHance® technology for enhanced tissue penetration and POTELLIGENT® technology to increase antibody-dependent cellular cytotoxicity (ADCC). Mechanistically, ARGX-111 binds with high affinity to c-Met and blocks both ligand-dependent (HGF-mediated) and ligand-independent activation of the receptor, thereby inhibiting downstream signaling pathways that promote cancer cell survival, proliferation, and metastasis. Additionally, it enhances immune-mediated killing of c-Met-expressing tumor cells via ADCC. Preclinical data also suggest depletion of MET-positive myeloid-derived suppressor cells in the tumor microenvironment. Clinical development has focused on advanced solid tumors overexpressing or amplified for c-Met[1][2][4][5][6].
