ARI-0001 (varnimcabtagene autoleucel) is an academic, autologous chimeric antigen receptor T-cell (CAR-T) therapy developed to target CD19-expressing B-cell malignancies. The therapy involves collecting a patient’s own T cells, genetically modifying them ex vivo with a lentiviral vector encoding a second-generation CAR construct containing an anti-CD19 single-chain variable fragment and a CD137 (4-1BB) costimulatory domain, and then reinfusing the engineered cells back into the patient. This approach enables the modified T cells to recognize and eliminate malignant B-cells expressing CD19. ARI-0001 was developed by Hospital Clínic de Barcelona/IDIBAPS in Spain and received hospital exemption approval from the Spanish Agency of Medicines and Medical Devices (AEMPS) for use in adults with relapsed or refractory acute lymphoblastic leukemia (ALL). Clinical trials have also evaluated its efficacy in chronic lymphocytic leukemia (CLL), Richter transformation, non-Hodgkin’s lymphoma, and other CD19+ malignancies[1][2][3][4][5][6][7].
