ARI0008 is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting the disialoganglioside GD2, a surface antigen highly expressed in diffuse intrinsic pontine glioma (DIPG) and other pediatric high-grade gliomas. Developed by the Fundació Sant Joan de Déu in collaboration with Hospital Clínic de Barcelona, ARI0008 is designed to address the critical need for effective treatments for H3K27M-mutant diffuse midline gliomas. The therapy involves the genetic modification of a patient's own T cells to express a CAR specific for GD2, which then recognizes and eliminates tumor cells. It is administered via both intravenous and intracerebroventricular routes to maximize exposure to the central nervous system and overcome the blood-brain barrier. ARI0008 is part of the academic ARI project, which focuses on developing accessible cell therapies within the public health system.
